Media Inoculation Instructions


Agar Plates: Use an inoculating loop to streak for isolated colonies. When applying A and P discs to the blood agar plates, place them in the heaviest area of inoculation. Incubate the plate at 37°C. for 18-24 hours.

Antimicrobial Sensitivity Plates: Using a cotton swab, inoculate the entire surface of the plate. Place discs on plate after swabbing. Incubate the plate at 37°C. for 18-24 hours.

Citrate, Lowenstein-Jensen, or Loeffler's Slants/Tubes: Using an inoculating loop, pick an isolated colony and streak the surface of the slant. Leave cap a little loose and incubate at 37°C. for 18-24 hours.

Urea Tube: Use an inoculating loop to pick an isolated colony and streak the surface of the slant. Tighten cap and incubate at 37°C. for 18-24 hours.

SIM Tube: Use an inoculating needle to pick an isolated colony and stab down the center of the butt all the way to the bottom. Incubate at 37°C. for 18-24 hours.

TSI Tube: Use an inoculating needle to pick an isolated colony and streak the surface of the slant, and then stab down the center of the butt all the way to the bottom. Incubate at 37°C. for 18-24 hours.
